About
Emmanuelle Bitoun is a leading molecular biologist whose research bridges the fundamental mechanisms of gene regulation with the pathogenesis of both childhood leukemia and neurodegenerative disease. Her work centers on the AF4 protein, a critical transcriptional cofactor that orchestrates RNA polymerase II elongation and chromatin remodeling. In a landmark 2006 study (308 citations), Bitoun provided the first evidence that the MLL fusion partner AF4 directly stimulates transcriptional elongation, a finding that reshaped our understanding of how mixed-lineage leukemia drives aggressive pediatric cancers. Beyond oncology, Bitoun made the striking discovery that AF4 also plays an essential role in the central nervous system. She demonstrated that mutations in Af4 cause the "robotic" mouse phenotype—characterized by severe ataxia and Purkinje cell loss—and identified Siah proteins as key mediators of AF4 function in the cerebellum. Her subsequent work revealed that AF4 is a critical regulator of the IGF-1 signaling pathway during Purkinje cell development, linking transcriptional dysregulation to a common pathological cascade in cerebellar ataxia. Through these contributions, Bitoun has illuminated a shared molecular vulnerability between leukemia and neurodegeneration, opening new avenues for therapeutic intervention.
