Protein Sequencing in the Post-Genomic Era

摘要
The availability, in recent years, of the nucleotide sequences of entire genomes, has radically reshaped approaches to protein sequencing. When the entire genomic sequence for an organism is known, it is a relatively simple matter to identify all possible open reading frames and to deduce the sequences of the proteins encoded by them. Improved mass spectrometric methods, coupled with robotic sampling techniques are revolutionizing the sequencing and identification of proteins. This article discusses the use of techniques, such as Matrix Assisted Laser Desorption Ionization (MALDI) and ElectroSpray Ionization (ESI), in protein sequencing and identification.
